Exact excerpt
The study screened a targeted sgRNA library for genes whose inactivation changes rare- versus common-codon-enriched fluorescent reporters. Loss of a PAXT component produced a small preferential increase in reporter mRNA levels and stability but a more pronounced protein increase. Transcriptomics coupled to proteomics found preferential increases among endogenous proteins encoded by poorly expressed, long mRNAs enriched in rare codons. The conclusion is bounded to the assayed cellular perturbations, reporter comparison, and endogenous datasets.
